Effects of single nucleotide polymorphisms in b-amylase1 alleles from barley on functional properties of the enzymes

چکیده
Differences in extractable b-amylase (1,4-a-D-glucan maltohydrolase, EC 3.2.1.2) activities exist between enzymes from malt and feed barleys (Hordeum vulgare L.). Others have documented few differences in the b-amylase (bmy1) gene coding regions and large differences in the non-coding regions of genomic DNA for endosperm specific b-amylases from malt and feed barleys. Researchers correlated differences in intron III with different activity levels in the seeds. Here we demonstrate that differences in the coding regions of bmy1 from a malt (‘Morex’) and a feed (‘Steptoe’) barley significantly affected the biochemical properties of the enzymes. We evaluated the contributions of the three single nucleotide polymorphisms that caused differences in the amino acid sequences at positions 115, 165, and 430. Two polymorphisms (positions 115 and 430) were found to impact the enzymatic activity of b-amylase. The effect of the single nucleotide polymorphism at position 165 was dependent upon the identity of the amino acid in position 115. The contributions of the three single nucleotide polymorphisms to b-amylase thermostability varied with assay temperature. These biochemical results explain the contribution of the favorable bmy1 allele from the feed barley Steptoe to the quantitative trait loci found on chromosome 4H at the bmy1 locus. © 2003 Published by Éditions scientifiques et médicales Elsevier SAS.

Influence of Vitamin D Receptor Gene Polymorphisms on Response to Pegylated Interferon in Chronic Hepatitis B Egyptian Patients
Background: We explored the effect of vitamin D receptor gene (VDR) polymorphisms in response to PEG-IFN treatment in Egyptian chronic hepatitis B (CHB) patients. Methods: Two hundred hepatitis B virus (HBV) patients (42.3±10.7 years) on PEG-IFN α-2a (180 μg /kg for 48 weeks) and one hundred control subjects (37.3 ±12 years) were enrolled in the study. Vitamin D levels a...
متن کاملInvestigation of GDF9 and BMP15 Polymorphisms in Mehraban Sheep to Find the Missenses as Impact on Protein
Utilization of fecundity genes such as GDF9 and BMP15 can help improve reproductive traits in sheep breeding programme. To evaluate effects of missense mutations on protein function, the polymorphisms of GDF9 and BMP15 genes were screened in twelve mehraban sheep using DNA sequencing, followed by protein structure modeling. Six single nucleotide polymorphism (SNPs) known as FecG mutations (G1-G...
متن کاملIn-silico study to identify the pathogenic single nucleotide polymorphisms in the coding region of CDKN2A gene
Background: CDKN2A, encoding two important tumor suppressor proteins p16 and p14, is a tumor suppressor gene. Mutations in this gene and subsequently the defect in p16 and p14 proteins lead to the downregulation of RB1/p53 and cancer malignancy. To identify the structural and functional effects of mutations, various powerful bioinformatics tools are available. The aim of this study is the ident...

متن کاملFREQUENCY OF C3435 MDR1 AND A6896G CYP3A5 SINGLE NUCLEOTIDE POLYMORPHISM IN AN IRANIAN POPULATION AND COMPARISON WITH OTHER ETHNIC GROUPS
ABSTRACT Background: It is well recognized that different patients respond in different ways to medications. The inter-individual variations are greater than the intera- individual variations, a finding consistent with the notion that inheritance is a determinant of drug responses. The recent identification of genetic polymorphisms in drug-metabolizing enzymes and drug transporters led to the ...
